The potato tuber moth (Phthorimaea operculella Zeller) (PTM) is a destructive pest. Bacillus thuringiensis cry genes encode crystal proteins that have toxic characteristics against PTM. aim of the present study was to develop PTM resistant cv. Agria using synthetic Bt cry1Ab gene. transformation vector constructed by inserting gene into pBI121 binary expression vector. Agrobacterium tumefaciens strain AGL1 harboring pBI35SCry1Ab construct used for transformation. results indicated out 97 putative transformed lines, 30 lines were PCR positive. Southern blot analysis revealed one three copies transgenes inserted transgenic lines. immunoassay test Cry protein. Furthermore, bioassays confirmed high mortality level neonate larvae. this suggested significant potential in pest control.
